Micro-preparative technique refers to the collection of factions after separation by Gas Chromatography to enhanced the concentration. The enrichment procedure is based on a repetitive adsorption of eluting compounds, after separation from a capillary column, by sucking them through an adsorption tube mounted on the outlet splitter. Optimization is easily performed during a supervised GC run in a semi automatic mode. Once optimized, repetitive injection/collections are performed using an autosampler. Full synchronization makes the system fully automated. The system directs the column effluent at any given time into the collecting system where the compounds of interest are adsorbed.
The target peaks were collected in different adsorption tubes based on retention times. The repeatedly enriched flavors or odorants can be dissolved and then used for structural measurements such as NMR. It provides important structural information for the analysis of flavor chemistry and the next step in synthesis.
The output of the column is split between the FID and the prep. The flow to the prep is stopped by an on/off valve. A vacuum pump is on the other side of the valve. When collecting the valve is opened, sucking the eluent from the column through the traps. At the end of the collection the valve is closed The carousel is advanced to the next tube.
